Abstract

Spiders represent a diverse and functionally important group of arthropods. They are important but poorly studied groups that play a significant role in regulation of insect and other invertebrate populations, and thus balancing the ecosystem. DNA extracted from spiders using the CTAB and GITC methods were compared for yield, quality, and effectiveness in PCR amplification. Here we present a modified protocol for DNA isolation and Polymerase Chain Reaction (PCR) conditions for Random Amplified Polymorphic DNA (RAPD) analysis of spiders based on a Guanidium Iso ThioCyanate (GITC) method. This technique is ideal for RAPD analysis of spider species. Reproducible amplification products were observed in all PCR reactions. The extraction method would allow the molecular analysis of DNA from different genera of spiders, species characterization and phylogenetic studies. © OmicsVista Group, All rights reserved.
